Hello,
My question is about the first step to make. Once you have a format (.MPEG2,...) , you want to wrap it in an MXF File. Does this format change with the KLV codage (since there will be Items separeted by Keys )?

The format of the essence does not change, but the stream of essence bytes may be broken up into multiple KLV packets.
